- Note : Discontinuation of treatment should be considered before age 11 for females and age 12 for males

- Therapy may cause increases in serum levels of testosterone, temporary increase in bone pain, ureteral obstruction and spinal cord compression, paralysis with or without fatal complications
- Periodically monitor serum testosterone and prostate specific antigen (PSA) levels during therapy
- Response to the drug should be monitored 1-2 months after the start of therapy with a GnRH stimulation test and sex steroid levels
- Events of hyperglycemia and an increased risk of developing diabetes have also been reported with the therapy. Monitor blood glucose and/or glycosylated hemoglobin (HbA1c) periodically in patients receiving a GnRH agonist and manage with current practice for treatment of hyperglycemia or diabetes
- Measurement of bone age for advancement should be done every 6-12 months
- Leuprolide may cause fetal harm when administered to a pregnant woman, hence it is contraindicated in women who are, or may become pregnant while receiving the drug
- Myocardial infarction, sudden cardiac death and stroke have been reported in association with use of GnRH agonists in men
- Carefully monitor and appropriately manage signs and symptoms suggestive of development of cardiovascular disease
- During the first few weeks of therapy, closely monitor patients with metastatic vertebral lesions and/or with urinary tract obstruction
- Convulsions reported in postmarketing studies among patients with a history of seizures, epilepsy, cerebrovascular disorders, CNS anomalies or tumors, and those taking concomitant medications that have been associated with convulsions such as bupropion and SSRIs

- Diabetes
- Depression
- Vertebral metastases (prostate CA use)
- Hematuria
