Adult Dosing
Endometriosis
- 400 mcg/day (200 mcg [1 spray] in one nostril in morning and 200 mcg [1 spray] in other nostril in evening)
- For long term suppression, 800 mcg/day may be necessary
- Max: 6 months
- Note: Start between days 2-4 of menstrual cycle
Pediatric Dosing
Central precocious puberty
- 1600 mcg/day (400 mcg [2 sprays] in each nostril bid). May be increased to 1800 mcg/day
- Continue until resumption of puberty desired
- Note: Allow 30 seconds to elapse between sprays

- Prior to initiating therapy, the diagnosis of central precocious puberty (CPP) must be established. Periodically monitor response as well as compliance in patients receiving the therapy at least for 6-8 weeks
- Ovarian cysts may occur in adult women with endometriosis in the first two months of the therapy
- Before starting treatment, pregnancy must be excluded because safe use of nafarelin acetate in pregnancy has not been established clinically
- Patients should use nonhormonal methods of contraception. Discontinue therapy if a patient becomes pregnant during therapy and apprise patient of the potential risk to the fetus
- Monitor bone mineral density if use > 6 months (endometriosis pts) or risk factors for decreased bone mineral content
- Symptoms of hypoestrogenism may also occur
- Elevations of phosphorus levels and eosinophil counts, and decreases in serum calcium levels and WBC counts have been documented
Cautions: Use cautiously in
- Polycystic ovary syndrome
- Osteoporosis
- Risk factors for osteoporosis
- Dyslipidemia
Supplemental Patient Information
- Patients should consult their physician they become pregnant during therapy

Breastfeeding: Safety unknown; due to the potential for serious adverse reactions in nursing infants, manufacturer recommends discontinuation of nursing or discontinuation of drug, taking into account the importance of the drug to the mother.
