Adjuvant treatment of early breast cancer
Extended adjuvant treatment of early breast cancer
- 2.5 mg PO qd
- Also in post-menopausal women, already treated with 5 yr of tamoxifen
Note:
- The treatment should be discontinued at tumor relapse
First line treatment of advanced breast cancer
Second-line treatment of advanced breast cancer
- 2.5 mg PO qd following antiestrogen therapy
Note:
- Continue the treatment until disease progression occurs
Pediatric Dosing
- Safety and effectiveness in pediatric patients have not been established

- Decreases in bone mineral density (BMD) may occur with use of therapy. Consider bone mineral density monitoring
- Decrease in lumbar spine and total hip bone mineral density (BMD) compared to baseline has occurred in patient receiving letrozole
- Patient receiving therapy has reported hypercholesterolemia compared to patients receiving tamoxifen. Consider monitoring of serum cholesterol during therapy
- Dose reduction is considered in patients with cirrhosis and/or severe hepatic function impairment
- Fatigue, dizziness and somnolence may occur with the use of therapy. Exercise caution when driving or using machinery
- Moderate decreases in lymphocyte counts, of uncertain clinical significance, were reported in some patients receiving therapy

Breastfeeding: It is unknown whether if letrozole is excreted in human milk. Manufacturer advises decision should be made whether to discontinue nursing or to discontinue the drug, taking into account the importance of the drug to the mother.
