Products

Amifostine is available as a lyophilized powder in single-dose vials containing 500 mg of amifostine on the anhydrous basis.1279 Reconstitute each vial with 9.7 mL of sodium chloride 0.9% to yield a solution containing amifostine 50 mg/mL.1279

The reconstituted solution should be further diluted in sodium chloride 0.9% to a final concentration of 5 to 40 mg/mL prior to administration.1279

Administration

Amifostine is administered by intravenous infusion.1279 When used as a chemoprotectant, the dose is administered by intravenous infusion over 15 minutes, starting 30 minutes prior to chemotherapy.1279 When used as a radioprotectant, the dose is administered by intravenous infusion over 3 minutes, starting 15 to 30 minutes prior to radiation therapy.1279

Stability

Amifostine lyophilized powder in intact vials should be stored at controlled room temperature.1279 Reconstituted solutions should be colorless and are stated to be chemically stable for 24 hours at 2 to 8°C or 5 hours at room temperature (approximately 25°C).1279

Diluted solutions for infusion prepared in PVC bags are stated to be stable for 24 hours at 2 to 8°C or 5 hours at room temperature (approximately 25°C).1279

Solutions should not be used if cloudiness or a precipitate is observed.1279 Any unused portions should be discarded.1279
