Linezolid is available as a single-use, ready-to-use solution for infusion in 100-, 200-, and 300-mL flexible plastic containers.3183; 3184 Each mL of linezolid (Zyvox, Pfizer) ready-to-use solution for infusion provides linezolid 2 mg along with dextrose, sodium citrate, and citric acid in water for injection.3183 Each mL of linezolid (Hospira) ready-to-use solution for infusion provides linezolid 2 mg along with citric acid anhydrous, sodium chloride, and sodium hydroxide in water for injection.3184 Sodium hydroxide and/or hydrochloric acid is used to adjust the pH.3183; 3184

Tonicity

Linezolid ready-to-use solutions for infusion are isotonic.3183; 3184

pH

Linezolid (Zyvox) ready-to-use solution for infusion is adjusted to pH 4.8.3183

Linezolid (Hospira) ready-to-use solution for infusion is adjusted to pH 4.4 to 5.2.3184

Sodium Content

The sodium concentration of linezolid (Zyvox) ready-to-use solution for infusion is 0.38 mg/mL or 1.7 mEq in a 100-mL bag, 3.3 mEq in a 200-mL bag, and 5 mEq in a 300-mL bag.3183

The sodium concentration of linezolid (Hospira) ready-to-use solution for infusion is 3.98 mg/mL or 52 mEq in a 300-mL bag.3184

Administration

Linezolid is administered by intravenous infusion over a period of 30 to 120 minutes.3183; 3184 If a common intravenous line is being used to administer other drugs in addition to linezolid, the line should be flushed with a compatible infusion solution (e.g., sodium chloride 0.9%, dextrose 5%, Ringer’s injection, lactated) prior to and following infusion of linezolid.3183; 3184

Stability

Linezolid solutions may exhibit a yellow color that can intensify over time without affecting the stability of the drug.3183; 3184 Intact containers should be kept in their protective overwrap until ready for use, and should be stored at controlled room temperature and protected from light and freezing.3183; 3184

Linezolid solutions should be visually inspected for particulate matter prior to administration.3183; 3184 The manufacturers state that additives should not be mixed with linezolid ready-to-use solution for infusion.3183; 3184

Temperature Effects

Linezolid (Pfizer) 2 mg/mL remained stable without noticeable degradation for 72 hours when stored at 70°C.3514

Sorption

Linezolid was found to be compatible with common types of intravenous administration sets including diethylhexyl phthalate (DEHP) plasticized PVC, trioctyl trimellitate (TOTM) plasticized PVC, and polyolefin sets. The total dose of linezolid was fully delivered with the delivered concentration remaining constant throughout. In addition, no detectable levels of plasticizer were found in the delivered solutions.2338

Additional Compatibility Information

Infusion Solutions

The manufacturers state that linezolid ready-to-use solutions for infusion are compatible with sodium chloride 0.9%, dextrose 5%, and Ringer’s injection, lactated.3183; 3184

One study found little to no loss of linezolid in Hartmann’s solution in 34 days at 25°C.3514

Peritoneal Dialysis Solutions

Linezolid 150, 300, and 600 mg/L in Dianeal PD-2 with 1.5 or 4.25% dextrose stored at 37, 25, and 4°C protected from light was physically compatible and exhibited no loss.2500
