Avelumab is available as a 20-mg/mL concentrate for injection in 10-mL preservative-free, single-dose vials containing 200 mg of the drug.3882 Each mL of the concentrate contains avelumab 20 mg with D-mannitol 51 mg, glacial acetic acid 0.6 mg, polysorbate 20 0.5 mg and sodium hydroxide 0.3 mg in water for injection.3882

The concentrate must be diluted prior to administration.3882 To prepare the drug for intravenous infusion, the appropriate dose of the concentrate should be withdrawn from the vial and added to an infusion bag containing 250 mL of sodium chloride 0.9% or sodium chloride 0.45%.3882 The solution should be mixed by gentle inversion and should not be shaken.3882 Any unused portion remaining in vials should be discarded.3882

Administration

Avelumab is administered by intravenous infusion over 60 minutes only after dilution in sodium chloride 0.9% or sodium chloride 0.45%.3882 Diluted solutions for infusion should be administered through a 0.2-µm low protein-binding inline filter.3882

Other drugs should not be co-administered with avelumab through the same infusion line.3882

Stability

Intact vials of avelumab should be stored at 2 to 8°C in the original carton to protect from light and should not be shaken or frozen.3882 The concentrate for injection is a clear, colorless to slightly yellow solution.3882 Vials should be visually inspected for particulate matter and discoloration prior to use; discard the solution if it is discolored, cloudy, or if particulate matter is present.3882

The manufacturer states diluted solutions should be protected from light and may be stored for no more than 4 hours at room temperature or 24 hours at 2 to 8°C.3882 Refrigerated solutions should be allowed to come to room temperature prior to administration.3882 Diluted solutions should not be frozen or shaken.3882

Filtration

Diluted solutions for infusion should be administered through a 0.2-µm low protein-binding inline filter.3882
