Primary hypercholesterolemia

Granules

• 5 g PO bid, may be increased q1-2 month up to 30 g/day divided qd-bid

Tablets

• 2 g PO bid, may be increased q1-2 month up to 16 g/day divided qd-bid

Hepatic Dose Adjustment

• Hepatic impairment: Dose adjustments not defined

• Do not administer therapy in its dry form to avoid accidental inhalation or esophageal distress. Always mix with water or other fluids before ingesting
• Secondary causes of hypercholesterolemia should be excluded, and a lipid profile performed to assess Total cholesterol, HDL-C, and triglycerides (TG) prior to initiating therapy
• Therapy may interfere with normal fat absorption and thus may reduce absorption of folic acid and fat soluble vitamins such as A, D, and K
• Vitamin K deficiency may lead to an increased bleeding tendency due to hypoprothrombinemia. In such cases use parenteral vitamin K1 and recurrences can be prevented by oral administration of vitamin K1
• Serum cholesterol and triglyceride levels should be determined periodically based on NCEP guidelines to confirm a favorable initial and adequate long-term response
• Colestipol may produce or severely worsen pre-existing constipation. Therefore increase dosage gradually in patients to minimize the risk of developing fecal impaction. Constipation associated with therapy may aggravate hemorrhoids
• Therapy may induce hypothyroidism in individuals with normal thyroid function
• Prolonged use may lead to the development of hyperchloremic acidosis
