Pharmacokinetics

Absorption: 80% absorbed after oral administration.

Distribution: Unknown.

Protein Binding: 97%.

Metabolism/Excretion: Extensively metabolized by the liver to inactive compounds. Converted intracellularly to at least two other antisecretory compounds.

Half-life: Children: 1.2–1.5 hr; Adults: 1.3–1.7 hr ( in geriatric patients and patients with impaired hepatic function).

Derm: cutaneous lupus erythematosus, rash.

F and E: hypomagnesemia (especially if treatment duration ge.gif3 mo).

GI: CLOSTRIDIOIDES DIFFICILE-ASSOCIATED DIARRHEA (CDAD), diarrhea, abdominal pain, fundic gland polyps, nausea.

GU: acute tubulointerstitial nephritis.

Hemat: vitamin B12 deficiency.

MS: bone fracture.

Neuro: dizziness, headache.

Misc: HYPERSENSITIVITY REACTIONS (INCLUDING ANAPHYLAXIS, ANGIOEDEMA, OR ACUTE TUBULOINTERSTITIAL NEPHRITIS), systemic lupus erythematosus.
